摘要:
目的 研究与环境致癌物活化、解毒有关的Ⅰ相和Ⅱ相代谢酶的mRNA水平与食管癌发病的关系。方法 收集93例淮安地区食管癌新发病例的食管癌组织及远端癌旁组织,应用SYBR Green Ⅰ实时荧光定量逆转录-聚合酶链反应(RT-PCR) 检测CYP2E1、GSTP1、MTHFR和NQO1等基因的mRNA水平;应用t检验和logistic回归分析评价基因的表达水平与食管癌的关系。结果 CYP2E1和MTHFR的mRNA水平在癌组织和配对癌旁组织之间有显著差异;条件logistic逐步回归分析结果显示CYP2E1、MTHFR表达水平异常与食管癌发病风险升高有关联[OR值(95%CI)分别为1.890 (1.439~2.481)和1.362 (1.107~1.677)]。结论 CYP2E1和MTHFR的表达水平改变与食管癌发病风险密切相关,在食管癌的高危人群筛查和早期诊断中具有重要意义。
Abstract:
Objective To investigate the relation between the mRNA expression levels of genes that involved in Ⅰ and Ⅱ phase of metabolism and activation of environmental carcinogens and the risk for ESCC. Methods Tumor and no-cancer tissues from 93 patients of newly diagnosed, untreated ESCC were collected in Huai'an County. The relative expression levels of four metabolism genes (CYP2E1, GSTP1, MTHFR and NQO1) were determined in cancer and no-cancer tissues by quantitative real-time RT-PCR using SYBR Green I. The expression of β-actin was used as an internal control. T-test and logistical regression analysis were used to test the differences of gene expression between cancer tissues and tissues adjacent to the tumors at a significance level of 5%.Results Compared with tissues adjacent to the tumors, the relative expression levels of CYP2E1 and MTHFR were significantly altered in esophageal cancer tissues. The conditional logistic regression analysis exhibited that the increased risk for esophageal cancer were significantly associated with aberrant expression of CYP2E1 and MTHFR [OR (95%CI)= 1.890 (1.439~2.481), OR (95%CI)=1.362 (1.107~1.677), respectively].Conclusion The increased expression of CYP2E1 and the reduced expression of MTHFR in esophagus were significantly associated with the risk for ESCC. Our data suggest an important etiology clue to CYP2E1 and MTHFR expression for the early progression of ESCC.
