摘要:
目的 建立以重组心肌肌钙蛋白I-C(cTnI-C)融合蛋白为包被抗原的间接酶联免疫吸附分析法(ELISA),检测人血循环中的抗cTnI自身抗体,为判断心肌损伤的预后提供依据。方法 采用自行构建表达的cTnI-C融合蛋白作为包被抗原,以棋盘法优化实验条件,建立检测人血清cTnI自身抗体的间接ELISA方法。采用建立的间接ELISA法在121例急性心肌梗死(AMI)中筛查抗cTnI自身抗体,并用蛋白印迹法(WB)进行确证。结果 121例AMI血清抗cTnI抗体的阳性率为10.74%,ELISA试验的灵敏度100%,特异性95.37%,Kappa值为0.815。结论 以cTnI-C融合蛋白作为包被蛋白建立的间接ELISA法具有良好的敏感度、特异性,可用于人血清抗cTnI自身抗体的筛查。
Abstract:
Objective To establish an indirect ELISA using cTnI-C recombinant protein as coating antigen in detecting cTnI autoantibody, and to provide with a method for detection of cTnI autoantibody in blood circulation and the prognostic test for myocardial damage. Methods cTnI-C recombinant protein was used as coating protein to establish indirect ELISA through checkerboard method. Screening of cTnI serum autoantibody in 121 AMI patients was done by indirect ELISA and was confirmed by western blotting. Results cTnI autoantibody was positively detected in 10.74% of the 121 AMI patients. The sensitivity of detecting cTnI autoantibody with this recombinant protein was 100% and specificity was 95%. Kappa value was 0.82. Conclusion The cTnI-C fusion protein can be used to indirect ELISA in detecting cTnI autoantibody.
