摘要:
目的 研究苗药防感香囊对小鼠肺组织Toll样受体7(TLR7)、干扰素α(IFNα)、干扰素β(IFNβ)的表达影响,从Ⅰ型干扰素信号通路角度探讨苗药防感香囊调节呼吸道免疫功能的作用机制,同时为苗药防感香囊有效预防呼吸道病毒感染提供科学理论依据。 方法 48只昆明种雄性小鼠随机分为空白对照组、香囊组、冷刺激组、香囊+冷刺激组,每组12只。香囊组、香囊+冷刺激组持续吸入苗药防感香囊30 d,第31天在已有干预基础之上给予冷刺激组、香囊+冷刺激组持续1周的冷刺激制造免疫低下模型,其后处死小鼠,留取标本检测。采用HE染色观察造模后肺组织结构,免疫组织化学染色、实时荧光PCR(Real time-PCR)、蛋白印迹法(Western blot)检测小鼠肺组织中TLR7、IFNα、IFNβ的基因/蛋白表达水平;采用酶联免疫吸附测定小鼠血清中IFNα、IFNβ分泌水平。 结果 与空白对照组比较,香囊组、香囊+冷刺激组小鼠肺组织中TLR7、IFNα/β的蛋白及mRNA表达增加(P<0.05),冷刺激组小鼠肺组织中TLR7、IFNα/β的蛋白及mRNA表达相对减少(P<0.05);与冷刺激组比较,香囊+冷刺激组小鼠肺组织中TLR7、IFNα/β的蛋白及mRNA表达水平增加(P<0.05);与空白对照组比较,仅香囊+冷刺激组中小鼠血清IFNα、IFNβ分泌水平升高(P<0.05),其他组间比较差异无统计学意义(P>0.05)。 结论 苗药防感香囊能够良性调节小鼠肺组织TLR7、IFNα、IFNβ基因和(或)蛋白表达水平;寒冷刺激能抑制小鼠肺组织TLR7、IFNα、IFNβ的转录与表达;苗药防感香囊能在一定程度上改善寒冷对小鼠肺组织TLR7、IFNα、IFNβ表达的抑制状态;苗药香囊可通过TLR7介导的通路中IFNα、IFNβ起到有效预防呼吸道感染,增强机体非特异免疫的作用。
Abstract:
Objective Respiratory tract infections are very common. Chinese medicine anti-allergic sachets have a good effect on adult and pediatric influenza, but the mechanism has not been widely reported. The study was aimed to investigate the effects of Miao Fanggan Sachets(MFS) on the expression of Toll-like receptor 7(TLR7), interferon alpha(IFNα)and interferon beta(IFNβ)in lung tissues of mice. Evaluate the preventive effects of MFS inhalation on the respiratory tract of mice and to explore the underlying molecular mechanism from the perspective of type I interferon pathway. Methods Konmin male mice (n1/4 48) of SPF grade, aged from 6-7 weeks, with a body mass between 25 g and 30 g were randomly divided into 4 groups consisting of normal control group (group A),inhalation sachet group (group B), cold stimulation group (group C), and sachet plus cold stimulation group (group D).Mice in group B, D were continuously given daily 10 grams of sachets for 12 hours and the sachet was changed every 6 days. From the 31st day, cold stimulation (Continual 7 days mice were exposed in 3±1℃ ice box 15 minutes a day) was given to group C to make mice immunocompromised model. After 37 days, mice were sacrificed under anesthesia and lungs harvested lung tissues. The expression levels of TLR7, IFNα, IFNβ was assessed using immunohistochemistry, real-time polymerase chain reaction (RT-PCR) and Western blot analyses. The secretory levels of IFNα, IFNβ in serum of mice were checked by ELISA. Results Compared to the blank control group, expressions of TLR7, IFNα, IFNβ in mice lung tissues were increased in Group B and D, there was statistically difference (P<0.05).Compared to the blank cold stimulation group, expressions of TLR7, IFNα, IFNβ in lung tissues of mice were increased in Miaoyao sachet group D(P<0.05).The expression of serum IFNα and IFNβ in the sachet plus cold stimulation group were higher than those in the control group (P<0.05).There was no significant difference between the other groups. Conclusion MFS up-regulates the expressions of genes and proteins of TLR7, IFNα and IFNβ in lung tissues of mice. Cold stimulation inhibits the transcription and expression of TLR7, IFNα and IFNβ in lung tissue of mice.MFS improves the inhibition of TLR7, IFNα and IFNβ expression in lung tissue of mice in cold condition to a certain extent and enhance local non-specific immune functions as well as prevent respiratory tract infection.
