摘要:
目的探讨Ybox结合蛋白1(YBX1)在结肠癌中的表达及其对结肠癌患者预后的影响,探索YBX1对结肠癌细胞迁移能力的影响。方法分析癌症基因组图谱(TCGA)数据库中YBX1在结肠癌中的mRNA表达水平。通过实时荧光定量PCR方法(qRTPCR)和蛋白质免疫印迹(Western blot)方法及免疫组化方法分别检测YBX1在结肠癌组织和细胞中的mRNA和蛋白表达量。67例结肠癌患者的肿瘤组织及癌旁组织经免疫组化染色方法分析YBX1蛋白表达,并分析患者临床特征和预后的关系,采用Cox比例风险回归模型分析结肠癌预后的影响因素。根据处理方式不同分为Mock组(用不含病毒的空载体溶液处理细胞)、NC组(无靶向作用的shRNA处理细胞)、LvYBX1组(过表达YBX1)、sh1YBX1组及sh2YBX1组(分别通过慢病毒载体携带的sh1YBX1 RNA、sh2YBX1 RNA,使细胞敲低YBX1基因的表达)。通过划痕实验与Transwell实验检测YBX1对结肠癌细胞迁移能力的影响。结果TCGA数据库样本分析显示,结肠腺癌YBX1的mRNA及蛋白表达水平显著高于癌旁组织(P<0.05)。qRTPCR分析显示,肿瘤组织中YBX1的mRNA表达水平显著高于邻近正常组织(P<0.01)。免疫组化结果及评分显示,YBX1在结肠癌组织中高表达(12.60±2.03),在癌旁组织中低表达(4.24±0.56),差异具有统计学意义(P<0.01)。KaplanMeier生存曲线结果显示,高表达YBX1的患者OS、DFS较低表达YBX1的患者明显降低(P<0.05)。在HCT15、HCT8细胞系中,与NC组相比,LvYBX1组的蛋白及mRNA水平明显升高(P<0.05)。在CACO2、SW480细胞系中,与NC组相比,sh1YBX1、sh2YBX1组的蛋白及mRNA水平明显降低(P<0.05)。与NC组相比,LvYBX1组HCT15与HCT8细胞的迁移能力均显著增强(P<0.05),sh1YBX1、sh2YBX1组CACO2与SW480细胞的迁移能力均显著降低(P<0.05)。结论YBX1高表达是结肠癌患者不良预后的危险因素,YBX1可作为结肠癌患者疾病进展及预后的潜在标志物。
Abstract:
ObjectiveThe study investigated the expression of Ybox binding protein 1 (YBX1) in colon cancer and its effect on the prognosis of colon cancer patients, and explored the effect of YBX1 on the migration ability of colon cancer cells. MethodsThe expression level of YBX1 mRNA in colorectal cancer was analyzed using the Cancer Genome Atlas (TCGA) database. The mRNA and protein expression of YBX1 in colon cancer tissues and cells were detected by realtime fluorescence quantitative PCR (qRTPCR) and protein immunoblotting (Western blot) methods and immunohistochemistry. The tumor tissues and adjacent cancer tissues of 67 colon cancer patients were analyzed for YBX1 protein expression using immunohistochemical staining. The relationship between patients′ clinical characteristics and prognosis was analyzed and Cox proportional hazards regression model was used to analyze prognostic factors of colorectal cancer. Cells were divided into four groups based on treatment methods: the Mock group (cells treated with an empty vector solution without virus), the NC group (cells treated with nontargeted shRNA), the LvYBX1 group (YBX1 overexpression group), the sh1YBX1 group (YBX1 knockdown group 1), and the sh2YBX1 group (YBX1 knockdown group 2). The effects of YBX1 on the migration ability of colon cancer cells were examined by scratch assay and Transwell assay.ResultsAnalysis of samples from the TCGA database revealed that mRNA and protein expression levels of YBX1 in colorectal adenocarcinoma were significantly higher than in adjacent noncancerous tissue. qRTPCR analysis showed that the mRNA expression level of YBX1 in tumor tissues was significantly higher than that in adjacent normal tissues (P<0.01). Immunohistochemical results and scoring showed that YBX1 was highly expressed in colon cancer tissues (12.60±2.03) and lowly expressed in adjacent normal tissues (4.24±0.56), with a statistically significant difference (P<0.01). KaplanMeier survival curve results showed that patients with high YBX1 expression had significantly lower overall survival (OS) and diseasefree survival (DFS) than those with low YBX1 expression (P<0.05). In the HCT15 and HCT8 cell lines, compared with the NC group, the protein and mRNA levels in the LvYBX1 group were significantly increased (P<0.05). In the CACO2 and SW480 cell lines, compared with the NC group, the protein and mRNA levels in the sh1YBX1 and sh2YBX1 groups were significantly decreased (P<0.05). Compared with the NC group, the migration ability of HCT15 and HCT8 cells in the LvYBX1 group was significantly enhanced (P<0.05), while the migration ability of CACO2 and SW480 cells in the sh1YBX1 and sh2YBX1 groups was significantly decreased (P<0.05).ConclusionHigh expression of YBX1 is a risk factor for poor prognosis in colorectal cancer patients, and YBX1 can serve as a potential biomarker for disease progression and prognosis in colorectal cancer patients.
