摘要:
目的 通过检测人类白细胞相关抗原-G(HLA-G)在子痫前期患者与正常胎盘组织中的表达,探讨HLA-G在子痫前期中的表达意义。方法 采用半定量逆转录-聚合酶链技术(RT-PCR)及免疫印迹(Western blot)分析技术分别检测了24例子痫前期患者(观察组,轻度子痫前期12例,重度子痫前期12例)及24例年龄相仿的正常晚期妊娠胎盘组织(对照组)中HLA-G的表达差异。结果 子痫前期患者胎盘组织中HLA-G基因与蛋白表达水平均显著低于正常妊娠组,两者比较差异有统计学意义(P<0.01),轻度与重度子痫前期比较差异无统计学意义(P>0.05)。结论 子痫前期胎盘组织HLA-G的表达明显下降,这一异常可能参与了子痫前期的发生和病理生理过程,但不能作为病情严重程度的筛选指标。
Abstract:
Objective To investigate the role of HLA-G in pre-eclampsia(PE) by examination of the expression of HLA-G in placentae from PE and normal pregnant. Methods The expression of HLA-G was detected in 24 cases of PE patients (experimental group, with 12 cases of mild PE and 12 cases of severe PE) and 24 cases of normal placentae in their late pregnancy (control group) using RT-PCR and Western Blotting respectively. Results Compared with control group, the levels of mRNA and protein of HLA-G in the placentae from patients with preeclampsia were significantly low (P<0.01).There was no significance in expression of HLA-G between mild and severe PE patients (P>0.05). Conclusion The expression level of HLA-G in placentae of PE patients decreased significantly. The abnormal expression of HLA-G might connect with the occurrence and pathology & physiology processes of PE, but can not used as a scanning factor for evaluation of the severity of the PE.
