摘要:
目的 探讨不同浓度的橄榄苦苷在不同作用时间对卵巢癌SKOV3细胞增殖的影响。方法 SKOV3卵巢癌细胞传代培养后接种于3块96孔板,每板实验组细胞用100、200、400、800 μg/mL的橄榄苦苷培养液,对照组细胞用McCoy′s 5A完全培养基。CCK-8检测法测定24、48、72h等3个时间点不同药物浓度的SKOV3细胞增殖情况。结果 相同作用时间下,与对照组比较,100、200、400、800 μg/mL的橄榄苦苷浓度对SKOV3细胞的增殖均有抑制作用,细胞生长A值随着药物浓度增大而减小(P<0
Abstract:
Objective To investigate the effects of different concentration of oleuropein on proliferation of ovarian cancer cell SKOV3 in different time.Methods SKOV3 ovarian cancer cells were inoculated in 3 blocks of 96 well plate. Cells of each trial group were treated with 100, 200, 400, 800 μg/mL oleuropein and vehicle, respectively. The proliferation of SKOV3 cells was determined by using CCK-8 assays after the treatment with indicated concentration of oleuropein for 24, 48, and 72 h.Results 100, 200, 400, and 800 μg/mL oleuropein significantly inhibited the proliferation of SKOV3 cells. Optical density (OD) was decreased accompanying with the dose increase of oleuropein (P<005). ODs of SKOV3 cells treated with 100, 200, and 400 μg/mL oleuropein were increased for the indicated incubation time (P<005). The proliferation of cells treated with 800 μg/mL oleuropein for 72 h was retarded compared with those for 24 h and 48 h (P<005). The inhibition rate of cell proliferation was increased with the dose increase of oleuropein treatment (P<005). However, there was no significant correlation with the incubation time (P>005).Conclusion Oleuropein could inhibit the proliferation of ovarian cancer cell SKOV3, the specific mechanisms needs further experimental study.
