摘要:
目的 通过观察不同浓度的胰岛素对视网膜Müller细胞生长活性的影响,探讨其在眼底病变中所起的作用。方法 运用组织块贴壁培养法培养兔视网膜Müller细胞,利用MTT法测定其在不同浓度胰岛素及作用时间下培养板中各孔的光吸收值。结果 当高浓度胰岛素作用时间≥3 d,光吸收(OD)值均明显下降,并随着浓度的增高及作用时间的延长而进一步减小。结论 当高浓度胰岛素作用达到一定时间时,对兔视网膜Müller细胞生长活性起着显著的抑制作用。
Abstract:
Objective To investage the role of Müller cells during the course of ocular fundus diseases by observe the effect of different concentrations of insulin on the proliferation of rabbit Müller cells. Methods Rabbit Müller cells were cultured by the explant tissue culture method. MTT assay was used to detect the OD value of 96-well plate containing different concentrations of insulin. Results The OD value was lower than that of the control group when the acting time of insulin was longer than three days, and was decreased with the increasing of insulin's concentration and acting time. Conclusion High concentration of insulin could significantly inhibit the proliferation of Müller cells when having appropriate acting time.
