摘要:
目的 研究慢性低氧高二氧化碳(CO2)环境下SD雌大鼠生殖器官发育的影响。 方法 30只SD雌鼠随机分为对照组和实验组,对照组雌鼠常温常压下喂养,实验组雌鼠每天暴露于低氧高CO2环境8 h,4周后与雄鼠合笼, 18 d后比较2组大鼠生殖器官脏器指数、生殖器官组织形态学,ELISA法检测大鼠血清血管内皮生长因子(VEGF)、雌二醇(E2)和孕激素受体(PR)水平差异。 结果 2组大鼠在生殖器官脏器指数、子宫内膜厚度、肌层厚度和腺体直径方面差异无统计学意义(P>0.05);实验组大鼠血清VEGF、E2和PR水平显著低于对照组[(13.44±1.88)ng/L vs (15.61±2.32)ng/L,(51.72±11.64)ng/L vs (68.78±9.91)ng/L,(1.09±0.13)ng/L vs (1.24±0.15)ng/L,P<0.05];实验组窦前卵泡数和卵泡总数显著少于对照组[(5.61±2.01)个 vs (9.10±4.85)个,(9.90±3.85)个 vs (12.10±5.05)个,P<0.05]);2组平均着床数比较差异无统计学意义(P>0.05),实验组活胎率显著少于对照组(72.9% vs 94.2%,P<0.05)。 结论 CO2高暴露未对雌性大鼠生殖器结构改变,但可显著影响卵泡生成能力并下调血清VEGF、E2和PR表达水平,导致孕鼠生育能力下降。
Abstract:
Objective To study the effects of chronic hypoxia and high carbon dioxide (CO2) on the development of reproductive organs in SD female rats. Methods Thirty SD female rats were randomly divided into control group and experimental group. Female rats in the control group were fed under normal temperature and pressure, while rats in the experimental group were exposed to low oxygen and high CO2 for 8 hours per day. The viscera index and reproductive organ morphology of two groups were compared. The expression levels of VEGF, E2 and PR in serum were determined by ELISA. Results There was no significant difference in visceral index and endometrial thickness, muscle thickness and gland diameter between two groups (P>0.05). The expression levels of serum VEGF, PR and E2 in the experimental group were significantly lower than those in the control group [(13.44±1.88)ng/L vs (15.61±2.32)ng/L,(51.72±11.64)ng/L vs (68.78±9.91)ng/L,(1.09±0.13)ng/L vs (1.24±0.15)ng/L,P<0.05]. The number of follicles and corpus luteum at both levels were significantly less in the experimental group than those in the control group [(5.61±2.01) vs (9.10±4.85),(9.90±3.85) vs (12.10±5.05),P<0.05]. There was no significant difference in the number of implantation of embryos between two groups. However, the number of live births in the experimental group was significantly less than that in the control group (72.9% vs 94.2%,P<0.05). Conclusion High CO2 exposure did not change the genital structure of female rats, but it could significantly affect the follicle generating ability and decrease expression of VEGF, E2 and PR, which leading to the reducing fertility in pregnant rats.
